Abstract
The oomycete Saprolegnia parasitica is a pathogen that infects mainly wild fish and cultivated fish, causing severe losses in the fish industry every year. Aquacultured fishes exists worldwide and constitutes an important food source for humans ever since overfishing started. Previously used methods for controlling pathogenic infections e.g. malachite green, has been proven non safe. It is today crucial to find an environmentally friendly method for controlling pathogenic infection such as saprolegniosis. One of six identified cellulose synthases in S. parasitica (SpCesA3.1) is highly expressed in the hyphal cell walls and therefore suggested as a potential target as drug control for saprolegniosis. To enable characterization of the structure and function of SpCesA3.1, it needs to be purified in its catalytically active full-lengths form. The aim of this project was to test a method for
heterologous expression, solubilization and purification of SpCesA3.1. The expected project outcome was an optimized protocol for the purification of membrane protein SpCesA3.1 in its catalytically active form. Solubilization and purification of SpCesA3.1 was assessed
experimentally, using detergent n-Dodecyl-B-D-Maltoside (DDM) as a solubilization agent and IMAC with preequilibrated Ni-NTA His•Bind® Resin protein purification. Experiments resulted in successful heterologous expressions in yeast strain LoGSA and FGY217,
transformed with pDD-8HIS-GFP-SpCesA3 plasmid. However, there was a negative purification of SpCesA3.1. Results were compared to previous research and a protocol on how to proceed with the optimization of solubilization and purification of SpCesA3.1 is presented here. It is proposed that the presented protocol is applied in further research on this subject.
Abstrakt
Oomyceten Saprolegnia parasitica är en fisk patogen som huvudsakligen infekterar vild fisk och odlad fisk, vilket orsakar allvarliga förluster för fiskindustrin varje år. Fiskodlingar finns över hela världen och utgör en av de viktigaste producenterna för fisk, ända sedan överfiske blev ett problem. Tidigare använda metoder för att kontrollera infektioner orsakade av patogen är malakitgrön, som har visat sig vara en hälsorisk. Det är idag avgörande att hitta miljövänliga metoder för att kontrollera saprolegniosis. En av sex identifierade
cellulosasyntaser i S. parasitica är SpCesA3.1, som uttrycks starkt i de hyfala cellväggarna och har därför föreslagits som ett potentiellt mål för läkemedelskontroll för saprolegniosis. För att kunna karakterisera struktur och funktion hos SpCesA3.1 måste proteinet renas fram i sin katalytiskt aktiva form. Målet med detta projekt var att testa en metod för att heterologt uttrycka, solubilisera och rena fram proteinet SpCesA3.1. Den förväntade slutprodukten var ett optimerat protokoll. Solubilisering och rening av SpCesA3.1 utvärderades experimentellt, där n-Dodecyl-B-D-Maltoside (DDM) användes som solubiliseringsmedel och IMAC för proteinrening. I detta projekt så lyckades SpCesA3.1 uttryckas heterologt i jäst celler från
Saccharomyces cerevisiae LoGSA och FGY217, men proteinet lyckades inte renas fram. Ett
protokoll med förslag på hur projektet ska fortgå presenteras här som en del av uppnådda slutsatser.
