Cloning, expression and
biochemical characterization of
pectinesterases from the oomycete
plant pathogen Phytophthora
infestans

ABSTRACT
Phytophthora infestans is a eukaryotic oomycete which infects mainly potato and tomato
causing late blight. Currently, the lack of an effective drug against this pathogen has resulted in large losses in annual world production of potatoesand tomatoes. P. Infestans infects through the direct germination of sporangia on the plant or the direct release of zoospore. Both forms lead to direct tissue penetration and development of mycelium within the host. In order to establis a successful intection, P. infestans secretes many proteins an enzymes. One of such protein is pectinesterase (PME) which is responsible for breaking down pectin, an important component in the plants cell wall. In the currently study, an in silico and in vitro analysis was performed for the characterization of this enzyme. In the in silico study and bioinformatics analysis, two pectinesterase proteins (PITG_01029 and PITG_02545) from by this pathogen were compared with other pectinesterases or similar proteins from 129 different species. In the in vitro study, one of the pectinesterase proteins, PITG_02545, was expressed in Escherichia coli and purified. After purification, the concentration of the pectinesterase was found to be high, approximately 4 mg/mL. The identity of the protein was also confirmed by mass spectrometry (MS). In addition, activity assay revealed that the purified enzyme was active. Future work is requiered to check the effect of inhibitors on the activity of this enzymes, that can further help in the development of anti-oomycetes drugs.

SAMMANFATTNING
Phytophthora infestans är en eukaryotisk oomycete som infekterar huvudsakligen potatis och
tomat som orsakar sen blight. För närvarande har avsaknaden av ett effektivt läkemedel mot denna patogen lett till stora förluster i den årliga världsproduktionen av potatis och tomater.
P. infestans infekterar genom direkt spiring av sporangia på växten eller genom direkt
frisättning av zoospore. Båda formerna leder till direkt vävnadsgenomträngning och utveckling av mycel i värden. For att upprätta en framgångsrik intektion utsöndar P. infestans många proteiner och enzymer. Ett av sådant protein är pektinesteras (PME) som ansvarar för att bryta ned pektin, en viktig komponent i växtens cellvägg. I den aktuella studien utfördes en in
silico- och in vitro-analys för karakterisering av detta enzym. I silikostudien och
bioinformatikanalys jämfördes två pektinesterasproteinerna, PITG_02545, i Escherichia coli och renades. Efter rening visade sig koncentrationen av pektinesteras vara hög, ungefär 4 mg/mL. Proteinets identitet bekräftades också med masspektrometri (MS). Dessutom avslöjade aktivitetsanalysen att det renade enzymet var aktivt. Framtida arbete krävs för att kontrollera effeckten av hämmare på aktiviteten hos dessa enzymer, som kan ytterligare hjälpa till i utvecklingen av läkemedel mot oomycetes.
