The antibody response in gene-targeted mice, producing IgM unable to activate complement

Title (Swedish)

Abstract

Comprehension about the immune system and how it is regulated is of major importance for fundamental immunology. The complement and immune complexes play an important role in autoimmune diseases. There are data showing that natural IgM is needed to mount a normal antibody response and it is suggested this lies in its ability to activate complement. Therefore, a knock-in mouse with a point mutation, making it unable to activate complement via IgM, was constructed. Characterization of this mutant mouse was done with several immunological methods and antibody responses were compared with normal mice.
