Purification and biochemical/immunological characterization of storage proteins from almond and
apple seed
Title (Swedish) Abstract
Food allergies against different fruits and nuts among humans are very common and can cause very severe allergic reactions and even be life threatening. One of the most common fruit allergies is that against apple which also has a near taxonomical relationship with almond. Storage proteins like 7S/11S Globulin and 2S Albumin are proteins that are common in seeds and nuts. These storage proteins are known to be proteins that cause severe allergic reactions, and are known as allergens. The aims of this project were to purify and characterize storage proteins from almond and apple seed with biochemical/immunological methods. The biochemical methods that were used during the project were preparative gel filtration, anion and cation exchange chromatography, SDS-PAGE and MS. The immunological characterization included analysis of IgE to the purified components in serum samples with known reactivity with storage proteins from other allergen sources from individuals with allergies against almond and/or apple The results indicate that new allergens from almond and apple seed have been found.
